I have a 16V automotive power supply feeding an Amp2, a 3B+, and the official 7" touch screen. I did a test power up and it worked fine. I hooked up the left speaker and did another test power up and nothing worked and SOMETHING got really hot. The Pi appears to be dead, and I don't know about the Amp2 and the screen.
Hope can I test the power regulation circuit on the Amp2 before I hook another Pi to it?
Could the 16V be the problem? Any ideas at all?

16V should be no problem. What exactly was this power supply? 16V doesn't sound like automative as these are usually using 12 or 24V.

I have it set up in Automotive Mode for 16V. It's configurable. Would 12V be better?
When I tested the input voltage on the Amp2 on the Pi, it was being pulled down to 1V and things were getting hot. I haven't tested it with ONLY the Amp2. Can I power up the Amp2 by itself without risk?
Yes, you can power the Amp2 alone.

I powered up the Amp2 by itself at 16V and it promptly let out the magic smoke:

this was either cause by over-voltage (much more than 16V) or wrong polarity. My guess is that you had the polarity reversed.

Neither of those things, however, were the case as measured by my meter in realtime.
It can be a short voltage peak that you don't see on your meter. A parts doesn't blow up just like this when running in its operating limits.

Provided the part is behaving correctly and not defective, yes.
The Amp2 board shipped with plastic standoffs, but I used brass standoffs to mount it to the Pi. Could that have grounded something that wasn't supposed to be? I didn't see any instructions in that regard.
As long as you didn't apply any voltage on the brass standoff, this shouldn't be a problem.
